Salt-inducible kinases (SIKs) are a family of serine/threonine protein kinases that belong to the AMP-activated protein kinase (AMPK) superfamily. They regulate diverse physiological processes, including metabolism, immune responses, circadian rhythms, and more. There are three main isoforms in mammals: SIK1, SIK2, and SIK3. They act as central mediators translating extracellular signals into transcriptional responses by phosphorylating substrates like CREB-regulated transcription coactivators (CRTCs) and class IIa histone deacetylases (HDAC4/5/7/9). SIKs are implicated in various diseases, including inflammatory diseases and cancer, making them potential therapeutic targets.
