Salt-inducible kinase 1 (SIK1) is a serine/threonine protein kinase and the founding member of the salt-inducible kinases, which are part of the AMP-activated protein kinase (AMPK) superfamily. SIK1 regulates multiple physiological processes, including transcriptional regulation, blood pressure homeostasis, and steroidogenesis. It is activated by LKB1-mediated phosphorylation and inhibited by PKA-mediated phosphorylation. Emerging evidence suggests that pan-SIK inhibition reduces pro-inflammatory cytokines, implicating therapeutic potential for inflammatory diseases.
