Overview

SAM and SH3 domain-containing protein 1 (SASH1) is an intracellular adaptor protein characterized by sterile alpha motif (SAM) and SRC homology 3 (SH3) domains. SASH1 functions as a tumor suppressor, inhibiting proliferation, invasion, and metastasis of cancer cells across multiple tissue types by negatively regulating pathways such as NF-κB and epithelial-to-mesenchymal transition. It also plays diverse roles in cell migration, cytoskeletal organization, pigmentation (especially melanocyte migration and melanogenesis), and vascular homeostasis. Loss or diminished expression of SASH1 is linked to a poor prognosis in several cancers and is causally associated with hereditary skin pigmentation disorders. SASH1 assembles molecular complexes by acting as a scaffold for signal transduction pathways but is not a classical druggable target like receptors or enzymes. No approved drugs currently act on SASH1 directly; however, its levels may serve as a useful biomarker for certain disease states.
