Overview

SAMD12 antisense RNA 1 (SAMD12-AS1) is a long non-coding RNA (lncRNA) located on chromosome 8, transcribed antisense to the SAMD12 gene. It is not translated into protein and acts as a regulatory RNA molecule in human cells. SAMD12-AS1 is upregulated in various tumor tissues, including liver and gastric cancers, as well as in neuroblastoma. Its expression is further enhanced by hepatitis B virus (HBV) infection through the viral HBx protein. SAMD12-AS1 promotes cancer cell proliferation and inhibits apoptosis primarily by interacting with nucleophosmin (NPM1), which affects the NPM1-HDM2-p53 pathway, resulting in reduced p53 protein stability and function. This activity contributes to tumorigenesis and malignant transformation by allowing cells to evade p53-mediated cell cycle arrest and apoptosis. SAMD12-AS1 is considered of interest both as a potential therapeutic target and as a biomarker for prognosis in p53-related cancers.
