Overview

Saponin adjuvants are amphiphilic plant-derived glycosides, commonly extracted from species such as Quillaja saponaria, that enhance vaccine-induced immune responses. Their immune-potentiating effects stem from their ability to promote robust activation of both innate and adaptive immunity, including the recruitment and stimulation of dendritic cells, cross-presentation of antigens, and induction of cytokine production. Purified saponin fractions such as QS-21 are included in licensed vaccines for COVID-19, malaria, and herpes zoster, substantially increasing the efficacy of these formulations. Saponins act by various mechanisms—delivery of antigens to APCs, activation of cytokine signaling, and upregulation of T-cell responses—although their precise molecular targets are not fully defined. Saponin adjuvants are notable for their strong ability to drive both humoral (antibody) and cellular (T-cell) responses, but attention must be given to their dose-related reactogenicity and safety profile.
