Sarbecovirus antigens are the structural proteins of the Sarbecovirus subgenus, which includes pathogens such as SARS-CoV and SARS-CoV-2. These antigens primarily consist of the spike (S), nucleocapsid (N), membrane (M), and envelope (E) proteins, which are essential for the virus's structural integrity and life cycle [3, 7]. The spike protein is the primary focus for drug and vaccine development as it mediates viral entry into host cells by binding to the angiotensin-converting enzyme 2 (ACE2) receptor [1, 5, 10]. Other antigens like the nucleocapsid protein are often used in diagnostic testing because of their relative abundance and high conservation [13, 15]. Therapeutic interventions, including monoclonal antibodies and vaccines, typically target specific epitopes on these antigens to neutralize the virus or elicit a protective immune response [2, 6, 8]. However, the rapid evolution of these viruses, particularly through mutations in the spike protein's receptor-binding domain, presents a significant challenge for maintaining long-term efficacy of these therapies [11, 14, 21].
Neutralization of viral entry by blocking receptor-binding domain interaction with ACE2, inhibition of viral-host membrane fusion, opsonization for immune clearance, and induction of protective humoral and cellular immunity [1, 6, 8, 23].
