The sarcoplasmic reticulum calcium release channel, primarily the ryanodine receptor (RyR), is a crucial ion channel in muscle cells responsible for releasing calcium ions from the sarcoplasmic reticulum (SR) into the cytoplasm. This calcium release initiates muscle contraction and is central to excitation-contraction coupling in skeletal, cardiac, and smooth muscle. There are three main isoforms: RyR1 (skeletal muscle), RyR2 (cardiac muscle), and RyR3 (less abundant, more broadly distributed). RyRs are large tetrameric complexes regulated by cytosolic calcium, accessory proteins, posttranslational modifications, and small molecules. Mutations in RyRs are linked to various diseases, including malignant hyperthermia and cardiac arrhythmias.
