Overview

The ORF1ab polyprotein is a large, multifunctional protein encoded by the open reading frame 1ab (ORF1ab) gene of SARS coronavirus 2 (SARS-CoV-2). This gene occupies about two-thirds of the viral genome and is essential for viral replication and transcription. The polyprotein is produced through a unique mechanism involving ribosomal frameshifting, resulting in two main products: pp1a and pp1ab. These are subsequently cleaved into 16 nonstructural proteins (nsps), which form the core machinery for viral RNA synthesis, processing, and modification. Key components derived from this polyprotein—especially RdRp and Mpro—are major drug targets in COVID‑19 therapy development.
