Overview

The N-terminal domain (NTD) is a structural component of the S1 subunit of the SARS coronavirus spike protein, playing a vital role in the early stages of viral infection. While the receptor-binding domain (RBD) is the primary site for interacting with the ACE2 receptor, the NTD facilitates initial viral attachment by binding to host cell surface molecules such as sialic acids, gangliosides, and co-receptors like AXL and DC-SIGN (1.3.4, 1.4.2). It is a major target for the host's neutralizing antibody response and contains a highly immunogenic region known as the NTD supersite (1.2.3, 1.4.2). Due to its role in immune recognition, the NTD is a hotspot for mutations that allow the virus to evade neutralizing antibodies, making it a critical focus for monitoring viral evolution (1.1.1, 1.4.1). In therapeutic development, monoclonal antibodies targeting the NTD are often used in combination with RBD-targeting agents to create synergistic cocktails that enhance neutralization and prevent mutational escape (1.1.3, 1.2.1). Beyond neutralization, some NTD-targeting antibodies can disrupt the integrity of the spike trimer or induce Fc-mediated effector functions to clear infected cells (1.2.2, 1.2.3).
