SARS-CoV-2 3C-like protease (3CLpro), also known as Mpro or main protease, is a cysteine protease essential for viral replication. It cleaves the two large polyproteins pp1a and pp1ab translated from the viral RNA genome at 11 conserved sites to release nonstructural proteins (nsps), which are crucial for viral transcription, replication, and recombination. The enzyme recognizes a specific cleavage sequence: Leu-Gln↓(Ser/Ala/Gly). Due to its essential role and the lack of close human homologs, it is a prime drug target for antiviral development.
