The SARS-CoV-2 3C-like proteinase (3CLpro), also known as main protease (Mpro), is a cysteine protease essential for processing viral polyproteins and thus for viral replication. It is a homodimer with a catalytic dyad composed of Cys145 and His41. Inhibition of 3CLpro prevents proper processing of viral proteins, blocking virus maturation and making it a key target for antiviral drug development against COVID-19.
