The SARS-CoV-2 nucleocapsid (N) protein is a highly conserved and abundant structural protein essential for viral RNA packaging and assembly. Peptides derived from the N protein are processed intracellularly and presented on the surface of infected cells by Major Histocompatibility Complex (MHC) molecules, specifically Human Leukocyte Antigens (HLA) in humans. These peptide-MHC (pMHC) complexes serve as critical targets for the cellular immune system, particularly CD8+ cytotoxic T cells. Because the N protein is more conserved across different variants than the Spike protein, these pMHC complexes are considered high-value targets for developing broad-spectrum vaccines and T-cell-based therapeutics. Recognition of these complexes by T-cell receptors (TCRs) triggers the destruction of infected cells and the release of antiviral cytokines like interferon-gamma. Therapeutic strategies targeting these complexes include TCR-engineered T-cell (TCR-T) therapies and multi-epitope vaccines designed to provide durable immunity against COVID-19.
