Overview

The SARS-CoV-2 spike protein N-terminal domain (NTD) is a critical component of the S1 subunit of the viral spike glycoprotein, specifically from the ancestral Wuhan-Hu-1 strain. While the receptor-binding domain (RBD) is the primary site for ACE2 interaction, the NTD plays a significant role in initial host cell attachment by binding to various co-receptors, such as sialic acids and heparan sulfate proteoglycans. It is a major antigenic site, and many potent neutralizing antibodies isolated from convalescent patients target this domain to inhibit viral entry. In the context of COVID-19, the NTD is a key focus for vaccine design and therapeutic antibody development. However, it is also a hotspot for mutations, which can lead to immune evasion and reduced susceptibility to existing treatments. Understanding the structural and functional properties of the Wuhan-Hu-1 NTD provides a baseline for monitoring the evolution of the virus and the efficacy of medical countermeasures.

Mechanism of action
