Overview

A satiating agent is a functional category of molecules, hormones, or pharmacological compounds that induce a feeling of fullness (satiety) and reduce food intake. These agents play a critical role in energy homeostasis by signaling the nutritional status of the body to the brain, primarily the hypothalamus and brainstem (physiology.org, 2007). Endogenous satiating agents include hormones such as glucagon-like peptide-1 (GLP-1), cholecystokinin (CCK), peptide YY (PYY), and amylin (nih.gov, 2021). In a therapeutic context, drugs that mimic or enhance the action of these agents, such as GLP-1 receptor agonists (e.g., semaglutide) and amylin analogs (e.g., pramlintide), are widely used to treat obesity and type 2 diabetes (ulster.ac.uk, 2023). Historically, specific substances like "satietin," a 50kDa glycopeptide found in serum, were proposed as primary satiating agents, though modern research focuses on the complex interplay of multiple gut-brain hormones (nih.gov, 1984). Because the term refers to a broad class of substances with diverse molecular targets rather than a single receptor or enzyme, it is considered a functional classification rather than a specific therapeutic target.

Mechanism of action

Satiating agents act by stimulating specific receptors in the central nervous system (e.g., GLP-1 receptor, Amylin receptor, Melanocortin 4 receptor) or the peripheral nervous system to signal fullness and reduce hunger. They often delay gastric emptying and modulate the release of other metabolic hormones.
