Scaffold attachment factor B1 (SAFB) is a multifunctional DNA-binding protein with specificity for scaffold or matrix attachment region (S/MAR) DNA elements. It is involved in chromatin organization by anchoring chromatin loops to the nuclear matrix and serves as a molecular base for assembling transcription complexes near actively transcribed genes. SAFB acts as a transcriptional corepressor, notably modulating estrogen receptor activity and negatively regulating cell proliferation. It has been implicated as a candidate tumor suppressor in breast cancer. SAFB does not function as a classical therapeutic receptor, enzyme, transporter, or direct drug target, but rather as a chromatin-associated regulatory protein impacting gene expression by modulating both DNA and protein interactions[1][2][4][3][6].
