Overview

Scaffold attachment factor B2 (SAFB2) is a multifunctional nuclear protein involved in the binding of scaffold/matrix attachment region DNA elements and is characterized as a DNA–RNA-binding protein (DRBP)[1][2]. It acts as a transcriptional corepressor, notably repressing the estrogen receptor alpha, and plays a role in regulating cell proliferation, the cell cycle, and RNA splicing[4][5][2][3]. SAFB2 directly interacts with chromatin and chromatin-modifying enzymes and is crucial for DNA repair and the cellular stress response[2][1][3]. It shows high expression in hormonally controlled tissues, particularly the male reproductive tract, where it regulates Sertoli cell functions and androgen receptor activity[3]. SAFB2 is implicated in cancer, especially breast cancer, but has few overt phenotypes when knocked out in mice, unlike its paralog SAFB1[3][5]. SAFB2 does not appear to be directly targeted by drugs, but it modulates pathways affected by established therapies through its repression of nuclear hormone receptor activity[5][4].
