Scaffolding protein involved in DNA repair (SPIDR/KIAA0146) is a scaffold protein essential for effective homologous recombination in human cells. SPIDR independently interacts with the Bloom syndrome helicase (BLM) and with RAD51, promoting the assembly and stability of BLM/RAD51 complexes at sites of DNA damage. Loss of SPIDR impairs DNA homologous recombination, increases the rate of sister chromatid exchange, and results in genome instability and cellular hypersensitivity to DNA-damaging agents[4][7]. SPIDR functions as a bridge, required for correct DNA repair choice and efficient maintenance of genomic integrity.
