Overview

Scavenger receptor class A member 5 (SCARA5) is a type II transmembrane glycoprotein belonging to the class A scavenger receptor family, which assembles as a cell-surface trimer and contains a collagen-like domain and a scavenger receptor cysteine-rich (SRCR) domain. It acts as an endocytic receptor for ferritin and von Willebrand factor, playing a key role in iron homeostasis, innate immune response, and clearance of plasma proteins. SCARA5 is expressed in various tissues, notably in epithelial cells and some endothelial cells, and functions as a tumor suppressor in several malignancies, where its downregulation is associated with increased proliferation, migration, and metastasis. Mechanistically, SCARA5 regulates cell cycle progression, apoptosis, unfolded protein response, and inhibits oncogenic signaling pathways such as PI3K/AKT. There are no known drugs directly targeting SCARA5, but its role as a tumor suppressor and plasma protein clearance receptor makes it a candidate for biomarker development and future therapeutic targeting.

Mechanism of action

As a tumor suppressor: SCARA5 overexpression inhibits proliferation and migration, induces apoptosis via inhibition of the PI3K/AKT pathway, and causes cell cycle arrest at G2/M phase. Endocytic receptor: SCARA5 enables uptake and clearance of ferritin and von Willebrand factor from plasma, potentially impacting iron and coagulation homeostasis.
