Overview

SCD divergent transcript (SCD-DT), also known as lncRNA RP11-34D15.2, is a long non-coding RNA identified as a key regulator of mitochondrial fatty acid metabolism and insulin sensitivity in pediatric obesity. It acts as a competitive endogenous RNA (ceRNA) by sponging microRNA-223 (miR-223), which leads to derepression (increased expression) of the transcriptional coactivator PGC-1α. This, in turn, stimulates FNDC5/irisin signaling, enhancing mitochondrial β-oxidation and free fatty acid clearance while improving insulin sensitivity. Levels of SCD-DT are significantly reduced in adipose tissue from obese children, suggesting a protective role against obesity-related insulin resistance and metabolic dysfunction. SCD-DT is not a receptor, enzyme, or transporter, but rather, a regulatory non-coding RNA with potential as a biomarker or therapeutic candidate in metabolic disease, but it is not yet considered a canonical drug target[1].
