Overview

Scleraxis (SCX) is a basic helix-loop-helix (bHLH) transcription factor critical for tendon and ligament development, maintenance, and repair[1][4][3]. It is expressed early in embryonic tendon progenitor cells and persists through tenocyte maturation[1][3]. SCX regulates the transcription of numerous extracellular matrix component genes necessary for tendon structure and function, and its loss results in severe hypoplasia or absence of force-transmitting tendons in animal models[1][4]. SCX function operates downstream of signaling pathways such as FGF and TGFβ during musculoskeletal development[3][4]. Although primarily a developmental biology marker and regulator, SCX's dysregulation has implications in congenital musculoskeletal disorders and tissue regeneration models, but there are currently no approved therapeutic drugs directly targeting SCX[1][4][3][6].
