A sclerosing agent is not a specific biological target or receptor, but rather a functional class of pharmacological substances used in sclerotherapy to induce the intentional destruction and subsequent fibrosis of blood vessels or other hollow structures. These agents are primarily utilized to treat conditions such as varicose veins, telangiectasias, hemorrhoids, and certain types of cysts or malformations. By causing immediate damage to the endothelial cell membrane or inducing osmotic stress, these substances trigger a coagulation cascade and inflammatory process that results in the replacement of the vessel lumen with a fibrous cord (NIH, 2022). Common examples include detergents like polidocanol and sodium tetradecyl sulfate, as well as osmotic agents like hypertonic saline. Because the term refers to a category of drugs rather than a molecular entity like an enzyme or receptor, it is classified as an incorrect target designation in a molecular context (StatPearls, 2023).
Sclerosing agents act by inducing direct chemical injury to the vascular endothelium or epithelial lining, leading to cell death and the exposure of subendothelial collagen. This triggers the coagulation cascade and a localized inflammatory response, resulting in the formation of a thrombus that eventually undergoes organization and fibrosis, effectively obliterating the lumen of the vessel or cavity (StatPearls, 2023; PubMed, PMID: 30725715).
