Sclerostin is a secreted glycoprotein that acts as a potent negative regulator of bone formation and plays a central role in bone homeostasis by inhibiting the Wnt/β-catenin signaling pathway, which is crucial for osteoblast function and bone growth.
Inhibition of Wnt/β-catenin signaling by binding to LRP5/6 co-receptors on osteoblasts, blocking canonical Wnt signaling required for new bone synthesis. Also binds to LRP4, acting as a molecular coordinator of both bone formation and resorption processes.
