Overview

Sclerostin domain-containing protein 1 (SOSTDC1) is a member of the sclerostin family, encoding a secreted, N-glycosylated protein with a C-terminal cystine knot-like domain. SOSTDC1 acts as a dual antagonist of BMP and Wnt signaling by directly binding BMP ligands and the LRP6 co-receptor, thereby regulating cellular proliferation, differentiation, programmed cell death, and tissue patterning during development. It is highly expressed in skin, intestine, bone, kidney, and vasculature. SOSTDC1 plays crucial roles in bone metabolism, tooth development, and is involved in cancer progression where its expression is frequently reduced. Genetic variants in SOSTDC1 are linked with bone density variation and supernumerary teeth. SOSTDC1’s epigenetic regulation and interactions with microRNAs further modulate its function in health and disease. At present, SOSTDC1 is considered a promising therapeutic target for bone disorders and various cancers.
