Scm-like with four MBT domains protein 1 (SFMBT1) is a chromatin-associated protein containing four malignant brain tumor (MBT) domains, homologous to members of the Polycomb group found in *Drosophila*[1][2][3]. SFMBT1 primarily acts as a transcriptional corepressor, modulating the expression of target genes through chromatin compaction and repression. It performs these functions as part of stable complexes with LSD1 and CoREST, both in somatic cells and during spermatogenesis. SFMBT1’s MBT domains bind histone tails and are essential for chromatin-binding, transcriptional repression, and the recruitment of histone-modifying complexes. It regulates genes involved in chromatin and nucleosome assembly, notably replication-dependent histone gene clusters, with roles in cell cycle progression and maintenance of undifferentiated myogenic progenitor cells through interaction with MYOD1. SFMBT1 is associated with bone sarcoma and other cancers. Currently, no drugs, biomarkers, or specific safety concerns are documented for SFMBT1 in clinical literature[1][2][3][4].
