Overview

SFMBT2 (Scm-like with four MBT domains protein 2) is a member of the Polycomb group (PcG) chromatin proteins, functioning as an epigenetic regulator primarily through maintenance of a repressive chromatin state. SFMBT2 is essential for stemness and maintenance of trophoblast progenitor cells in the placenta. It mediates transcriptional repression by binding to chromocenters, LINE elements, and major satellite sequences, and interacts with repressive histone marks such as H3K9me2, H4K20me2, and mono-ubiquitinated H2AK119. In cancer, especially prostate cancer, SFMBT2 acts as a negative regulator of cell migration and invasion, partly via repression of matrix metalloproteinases (MMPs) and associated cofactors such as YY1, RNF2, N-CoR, and HDAC1/3. Reduced expression of SFMBT2 correlates with higher metastatic potential in prostate cancer cells, suggesting a tumor suppressor-like role. There are no approved therapies targeting SFMBT2 directly, but its expression and regulatory networks are of interest for future therapeutic and biomarker development.
