Overview

Sebum production regulators in sebocytes refer to a collective group of receptors, enzymes, and transcription factors that govern the synthesis and secretion of lipids by sebaceous glands (Zouboulis et al., 2004, PMID: 15556719). Central to this group is the androgen receptor (AR), which, upon activation by dihydrotestosterone, promotes sebocyte proliferation and lipid production (Makrantonaki et al., 2011, PMID: 21272114). Other critical regulators include peroxisome proliferator-activated receptors (PPARs), particularly PPAR-gamma, and the melanocortin-5 receptor (MC5R), which play essential roles in sebocyte differentiation and lipogenesis (Clayton et al., 2020, PMID: 31833161). Dysregulation of these regulatory networks is a hallmark of acne vulgaris and seborrhea, leading to hyperseborrhea and subsequent follicular inflammation. Therapeutic strategies targeting these regulators include topical androgen antagonists like clascoterone and systemic retinoids like isotretinoin, which induce sebocyte apoptosis and significantly reduce sebum output (Layton, 2009, PMID: 19468332).
