Overview

SEC14-like protein 2 (SEC14L2) is a cytosolic carrier protein and member of the lipid-binding SEC14 domain family. It binds hydrophobic molecules, notably alpha-tocopherol (vitamin E), with high affinity, and is involved in the cellular uptake and intracellular transport of vitamin E and its derivatives[1][3][6]. SEC14L2 stimulates squalene monooxygenase, a key enzyme in the cholesterol biosynthetic pathway, and binds squalene, implicating it in the regulation of cholesterol metabolism[3][5][6]. This protein modulates signaling pathways by enhancing vitamin E-mediated protection against lipid peroxidation, regulating the availability of phosphoinositides, and integrating signals through phospholipase C, PIP2 turnover, and intracellular calcium release[1][7]. It acts as a host factor that enhances hepatitis C virus (HCV) replication by protecting cells from oxidative damage[1]. SEC14L2 is linked to human disease susceptibility, including neurodegenerative disorders, cancer, and inherited vitamin E deficiency[2][3].
