SEC31 homolog A, COPII coat complex component (SEC31A), is a structural component of the coat protein complex II (COPII) that regulates vesicle budding from the endoplasmic reticulum (ER) to the Golgi apparatus. SEC31A, a close functional homolog of yeast Sec31p, constitutes part of the outer shell of the COPII coat. It engages in both membrane deformation and cargo selection during ER vesicle formation, working together with other COPII subunits (Sar1, Sec23/24, Sec13) for efficient ER-to-Golgi transport of secretory proteins. In mammalian cells, SEC31A contains WD40 repeats and a proline-rich domain, interacts with SEC13, is regulated by monoubiquitylation (mediated by CUL3-KLHL12 to modulate vesicle size), and its membrane association is influenced by calcium-dependent ALG-2 binding. Mutations or dysregulation of SEC31A have been linked to rare syndromic diseases but it is not currently regarded as a direct therapeutic target for drugs[1][2][3][4][6].
