The Sec61 translocon is a highly conserved, heterotrimeric protein complex that forms the core of the protein-conducting channel in the endoplasmic reticulum (ER) membrane of eukaryotes and in the plasma membrane of prokaryotes. Its primary function is to mediate the translocation or integration of newly synthesized polypeptides into or across membranes, playing a central role in secretory and membrane protein biogenesis. Beyond its role in protein translocation, Sec61 also functions as an ER calcium leak channel, making it relevant to various disease states.
