Overview

SEC62 preprotein translocation factor (SEC62) is an integral membrane protein of the endoplasmic reticulum that, together with SEC63 and SEC61, forms part of the central protein translocation machinery in eukaryotic cells[1][2][3][5]. It mediates post-translational transport of specific precursor polypeptides into the ER, participates in ER-phagy (the degradation of excess ER and associated proteins after ER stress), and is involved in the maintenance of calcium homeostasis through interactions with calmodulin[1][2][4][5]. SEC62 is frequently overexpressed in multiple cancer types, correlating with poor prognosis, enhanced migration/invasion, and increased stress tolerance of tumor cells[1][2]. Its expression serves as a predictive and prognostic biomarker in various cancers, and it represents a potential therapeutic target—for example, drugs such as trifluoperazine (a calmodulin antagonist) can antagonize SEC62-driven phenotypes[2]. SEC62 is also implicated in cellular recovery from ER stress and interacts with core ER stress and protein quality control pathways, such as the unfolded protein response (UPR) and ER-associated degradation (ERAD)[1][2][5].
