SMAC is a mitochondrial protein that promotes apoptosis by antagonizing the inhibitory effects of IAPs on caspases. Upon apoptotic stimuli, SMAC is released into the cytosol and binds to IAPs, facilitating caspase activation and cell death. SMAC mimetics are being developed as cancer therapeutics to enhance tumor cell susceptibility to apoptosis.
SMAC mimetics bind to IAPs, neutralizing their inhibitory effect on caspases, thereby promoting apoptosis.
