Secretogranin V (SCG5), also known as neuroendocrine protein 7B2, is a secreted chaperone protein highly expressed in neuroendocrine tissues. Its main biological function is to prevent the aggregation of secreted proteins, notably including those related to neurodegenerative and metabolic disorders. SCG5 acts as a chaperone for prohormone convertase PC2, facilitating its transport and activation in the secretory pathway, and it also participates in pituitary hormone regulation. Importantly, SCG5/7B2 demonstrates anti-aggregation properties against amyloid-β and α-synuclein, thus providing cellular protection against neurotoxicity. In certain cancers, such as medulloblastoma, epigenetic silencing of the SCG5 gene is linked to enhanced tumor cell proliferation, suggesting a tumor suppressor role. SCG5 belongs to the granin family and is considered multifunctional, critical for maintaining neural and endocrine integrity[1][2][3].
