Secretory calcium-binding phosphoprotein proline-glutamine rich 1 (SCPPPQ1) is a small, secreted protein localized in the basal lamina of the cell-tooth interface, particularly in tooth and junctional epithelial tissues[2][6]. It is highly conserved, proline- and glutamine-rich, and encoded within the secretory calcium-binding phosphoprotein (SCPP) gene cluster[2]. SCPPPQ1 plays a structural role in mediating the attachment of epithelial cells to mineralized tooth surfaces, helping maintain the integrity of the gingival seal that protects against bacterial invasion[2][6]. Notably, SCPPPQ1 shows resistance to bacterial degradation and exhibits antimicrobial properties, acting bactericidally against oral pathogens such as *Porphyromonas gingivalis* without being toxic to mammalian cells[1]. There is no direct evidence it is a therapeutic target, drug target, or biomarker in clinical development, but its unique properties indicate a role in oral health and as a component of the extracellular matrix[1][2][6].
