Overview

Secretory carrier membrane protein 5 (SCAMP5) is a tetraspan integral membrane protein primarily involved in membrane trafficking within secretory and endocytic organelles[1][2][3]. It is a member of the SCAMP family, characterized by four transmembrane domains and cytoplasmic N- and C- termini[2]. SCAMP5 is distinct among SCAMPs for its restricted expression pattern: it is most abundant in neuronal cells of the central nervous system and in human immune plasmacytoid dendritic cells (pDCs), where it closely associates with the Golgi apparatus and the interferon (IFN) secretion pathway[1]. SCAMP5 regulates vesicular trafficking, protein sorting, and the proper surface expression of certain ion channels, especially T-type calcium channels, and is implicated in synaptic vesicle physiology and the immune response[3]. Genetic variants and mutations in SCAMP5 have been associated with neurological disorders (including neurodevelopmental delay, epilepsy, and Parkinson’s disease) and autoimmune conditions such as systemic lupus erythematosus, highlighting its biological and clinical significance[1][3].
