Overview

SEH1-like nucleoporin (SEH1L) is a scaffold protein that is a core component of the nuclear pore complex (NPC), specifically part of the Nup107-160 subcomplex essential for nuclear-cytoplasmic transport, mitotic spindle formation, and chromosome segregation[1][3][5]. SEH1L also participates in the GATOR2 complex, which regulates the amino acid-sensing mTORC1 signaling pathway, impacting cell growth, proliferation, and differentiation[2][3][4]. Through its roles in both the nuclear pore and GATOR2, SEH1L coordinates multiple aspects of cellular physiology, including cell division, nutrient signaling, and brain development, although it is not considered a direct therapeutic drug target. SEH1L dysfunction has been linked to specific diseases such as congenital nail abnormalities and has regulatory connections to cancer and neurodevelopmental disorders via the mTORC1 pathway[2][3][4]. The protein is highly conserved across eukaryotes and has structural similarity to yeast Seh1 and Sec13[3][5].
