Overview

Seizure related 6 homolog (SEZ6) is a type I transmembrane protein highly expressed in the nervous system, critical for neuronal development, synaptic connectivity, and motor coordination[1][3]. It acts as a trafficking factor for kainate receptor subunits GluK2/3, regulating their glycosylation and surface localization in neurons, and is required for the modification of these receptors with specific glycan structures[1]. SEZ6 is a major substrate of the β-secretase enzyme BACE1 in the brain, and changes in SEZ6 processing or function have been implicated in epilepsy, Alzheimer's disease, and other psychiatric or neurodegenerative disorders[1][3][4]. It is characterized by three CUB domains, five SCR domains, and potential roles in neuronal membrane signaling and cell-cell recognition. The soluble ectodomain shed from SEZ6 can be detected in cerebrospinal fluid and may serve as a biomarker for monitoring BACE1 activity in neurological disease research[3][4].
