Seizure-related homolog 6 protein (SEZ6) is a type I transmembrane glycoprotein highly expressed in neurons. It plays critical roles in neuronal development, synaptic regulation, and complement system regulation. SEZ6 influences synapse numbers, dendritic morphology, neuronal connectivity, and learning/memory processes. It also regulates glycosylation and trafficking of kainate receptor subunits GluK2/GluK3. Furthermore, SEZ6 inhibits opsonization by classical/alternative complement pathways. It is linked to Alzheimer's disease, schizophrenia, autism spectrum disorders, and is overexpressed in small cell lung cancer, making it a target for antibody-drug conjugates.
