Semaphorin 3F antisense RNA 1 (SEMA3F-AS1)

Target
SEMA3F-AS1
Molecular classification
Long noncoding RNA (lncRNA), Antisense RNA

Overview

Semaphorin 3F antisense RNA 1 (SEMA3F-AS1) is a long noncoding antisense RNA transcribed opposite the SEMA3F gene. Its primary biological role is the regulation of SEMA3F expression, influencing processes such as cell proliferation, migration, invasion, and potentially apoptosis and angiogenesis. SEMA3F-AS1 can act through chromatin remodeling and epigenetic mechanisms, such as histone modification, to modulate expression of tumor suppressors or oncogenes. Dysfunction or aberrant expression of SEMA3F-AS1 is implicated in several cancers, with altered levels serving as diagnostic or prognostic biomarkers. While not a receptor, enzyme, or transporter, SEMA3F-AS1 is actively investigated as a therapeutic target and biomarker in oncology[3][4][5].

Mechanism of action

Epigenetic regulation of target gene expression via recruitment of chromatin remodeling complexes; Histone modification (e.g., influencing H3K27 methylation or H3K9 acetylation of target gene promoter regions[3]); Antisense inhibition or upregulation of cognate gene (SEMA3F, RBM5) with downstream effects on cell cycle, proliferation, invasion, and apoptosis[3][4]

03

Biological functions

Gene expression regulationEpigenetic modulation (chromatin remodeling, histone modification)Modulation of cell proliferation, migration, and invasion (based on analogous lncRNA mechanisms[4][5])May regulate apoptosis and angiogenesis through modulation of cognate protein-coding gene expression[3][5]
04

Disease associations

Cancer (notably colorectal carcinoma, osteosarcoma)[3][4][5]Potential role in other malignancies driven by dysregulation of SEMA3F
05

Safety considerations

Potential off-target effects and regulatory complexity for therapeutic modulationUnknown long-term safety as lncRNA-based targets are still investigational
06

Biomarkers

SEMA3F-AS1 upregulation or downregulation as a biomarker in tumor tissue (e.g., CRC, Os)Downstream effects on SEMA3F or RBM5 expression levels in specific cancers[3][4]
