Overview

Semaphorin-3G is a secreted member of the class 3 semaphorin protein family, characterized by the presence of a sema domain and functioning primarily as a cell guidance cue in neuronal development and regulation of angiogenesis. SEMA3G binds neuropilin-2 (NRP2) to initiate signaling via associated plexin receptors, affecting vascular and neuronal cell behaviors by regulating migration, proliferation, and cell adhesion. In the vasculature, SEMA3G acts as an autocrine signal for endothelial cells and as a paracrine regulator for smooth muscle cells, with context-dependent pro- or anti-angiogenic activity. Its loss is associated with vascular instability and disease, and it plays additional roles in neural function, adipogenesis, and tumor suppression.
