Semaphorin-4G is a transmembrane protein belonging to the semaphorin family, characterized by a sema domain and an immunoglobulin domain in its extracellular portion. It acts as a cell surface receptor, most notably for plexin-B2 (PLXNB2), and is involved in critical biological processes such as axon guidance, immune cell maintenance, vascular growth, and regulation of cell motility. In mammals, Semaphorin-4G functions as a guidance ligand via multimeric receptor complexes, often involving plexin proteins. It is encoded by the SEMA4G gene on chromosome 10q24.31 and is implicated in developmental processes of the nervous system (for example, development of cerebellar granule cells) and in cancer biology, where it may have oncogenic or tumor-suppressive roles that are type-specific. Inherited or acquired variations in this gene have been associated with neurodevelopmental disorders, including autism spectrum disorder. No drugs are currently known to directly target Semaphorin-4G, and its mechanism as a therapeutic target remains to be fully defined.
