Overview

Semenogelin-2 is a secreted protein encoded by the SEMG2 gene in humans and is a major constituent of human seminal fluid[1][3][4][8]. It primarily functions in the formation of a gel matrix (sperm coagulum) that encases the ejaculated spermatozoa, regulating timing and release for subsequent sperm motility after liquefaction by prostate-specific antigen (PSA)-mediated proteolysis[1][3][8]. Alongside semenogelin-1, SEMG2 ensures initial immobilization and later release of sperm, and products of its breakdown contribute to antibacterial protection[2][3]. SEMG2 belongs to the class of cancer-testis antigens (non-X linked CTA) and is normally expressed only in male reproductive tissues but has been observed to be re-expressed in several cancers, making it a potential tumor biomarker[2]. There is currently little evidence of direct pharmacological targeting or therapeutic modulation of SEMG2, and its exact functional role in non-reproductive tissues and cancer biology remains under investigation[2].
