Overview

Senescence associated long non-coding RNA 1 (SALRNA1, also called SAL-RNA1 or XLOC_023166) is a long non-coding RNA identified in human cells, notably in type II alveolar epithelial cells. SALRNA1 is downregulated in cellular senescence and functions to modulate the SIRT1-FOXO3a/p53 signaling pathway, thereby reducing cellular senescence induced by stressors such as cigarette smoke. Experimental work suggests that upregulation or overexpression of SALRNA1 can reduce markers of senescence and apoptosis in lung epithelial cells, implicating it as a potential therapeutic target to ameliorate pathological aging processes, especially in lung diseases like COPD. SALRNA1 is considered part of the broader lncRNA family, a class of RNA molecules involved in epigenetic regulation and control of gene expression, with emerging roles in various disease pathologies

Mechanism of action

Modulation of SIRT1 signaling (SALRNA1 overexpression leads to upregulation of SIRT1, which decreases cellular senescence via regulation of FOXO3a and p53 transcription)

03

Biological functions

Regulation of cellular senescenceRegulation of gene expression (notably through the SIRT1-FOXO3a/p53 pathway)Modulation of apoptosis in lung epithelial cellsPutative roles in genomic stability and developmentPotential involvement in oxidative stress responses
04

Disease associations

Chronic obstructive pulmonary disease (COPD)Cellular aging/senescence pathwaysOther diseases associated with impaired cellular senescence (possible links to cancer, diabetes, neurodegenerative disease suggested for lncRNAs in general, but not specifically proven for SALRNA1)
05

Safety considerations

Not established; as a non-coding RNA target, off-target effects from lncRNA modulation or gene therapy delivery methods would be general concerns, but there are no specific clinical safety data for SALRNA1 targeting yet
