Overview

Sensor histidine kinase ComD is a membrane-bound receptor in Streptococcus mutans that serves as the primary sensor for the competence-stimulating peptide (CSP) in a density-dependent quorum-sensing system [1, 3]. As part of the ComCDE two-component system, ComD undergoes autophosphorylation upon CSP binding and subsequently activates its cognate response regulator, ComE, to initiate a signaling cascade [4, 8]. This pathway is a master regulator of S. mutans virulence, controlling the transition from a planktonic state to a robust biofilm (dental plaque), as well as regulating acid tolerance and the production of mutacins [6, 11]. Because S. mutans is the principal etiological agent of dental caries, ComD is a high-priority therapeutic target for developing anti-biofilm agents that attenuate bacterial pathogenicity without necessarily exerting the broad-spectrum selective pressure of traditional antibiotics [8, 12]. Experimental strategies targeting ComD include the use of CSP analogs and small-molecule inhibitors to disrupt bacterial communication and prevent the formation of cariogenic plaques [1, 15].

Mechanism of action

Inhibition of the ComD sensor histidine kinase prevents the activation of the ComDE two-component system, thereby suppressing the expression of genes involved in biofilm formation, acid production, and genetic competence.
