Overview

Seoul virus glycoprotein Gn is a critical surface component of the Seoul orthohantavirus, a member of the Hantaviridae family [2, 13]. It is encoded by the M (medium) segment of the viral genome as part of a precursor polyprotein that is co-translationally cleaved into Gn and Gc subunits [3, 12]. These glycoproteins form heterodimeric spikes on the viral envelope, which are essential for attachment to host cell receptors, such as beta-3 integrins, and subsequent clathrin-mediated endocytosis [2, 5]. Gn also plays a vital role in viral assembly by interacting with the nucleocapsid protein and directing the budding process at the Golgi apparatus [5, 13]. In humans, Seoul virus infection can lead to hemorrhagic fever with renal syndrome (HFRS), characterized by fever, acute kidney injury, and vascular leakage [3, 6]. While there are currently no FDA-approved vaccines or targeted therapeutics specifically for Seoul virus, glycoprotein Gn is a primary target for the development of neutralizing monoclonal antibodies and entry-inhibiting antiviral drugs [4, 12]. Its surface exposure and essential role in the viral life cycle make it a focal point for diagnostic assays and therapeutic intervention strategies [12, 15].
