Overview

Septin 7 (SEPT7) is a pivotal member of the septin family, a group of GTP-binding proteins that assemble into filaments and rings to provide structural support and act as molecular scaffolds within the cell [1, 2]. As the only member of the Septin Group 7, it is considered the essential core subunit required for the formation and stability of most septin hetero-oligomeric complexes; its depletion typically leads to the degradation of other septin proteins [3, 4]. Biologically, SEPT7 is crucial for cytokinesis, where it facilitates the final stages of cell division, and it also plays significant roles in cell polarity, ciliogenesis, and dendritic spine morphogenesis in neurons [1, 4]. In clinical contexts, SEPT7 dysregulation is associated with various cancers, including gliomas and leukemias, as well as neurodevelopmental and psychiatric disorders like schizophrenia [5, 6]. While traditional small-molecule inhibitors like forchlorfenuron target the protein's assembly, the targeting of SEPT7 mRNA via RNA interference (siRNA) or antisense oligonucleotides (ASOs) represents a specific therapeutic approach to modulate its expression in hyperproliferative or pathological states [7, 8].
