Overview

Sequestosome-1 (p62) is a multifunctional scaffold/adaptor protein involved in selective autophagy and the regulation of signaling pathways such as NF-kB and Nrf2. It acts as a cargo receptor for ubiquitinated proteins, facilitating their degradation via autophagy and the ubiquitin-proteasome system. p62 also plays a role in bone remodeling and is implicated in diseases such as Paget’s disease of bone, ALS, and FTLD. Its diverse functions are mediated by multiple functional domains, including PB1, ZZ-type zinc finger, TRAF6-binding domain, LIR/KIR motifs, and UBA.
