Serine peptidase inhibitor Kazal type 14 (SPINK14) is a protein-coding gene proposed to function as a serine protease inhibitor belonging to the Kazal-type family. The protein is likely to reside in the extracellular space, where it may regulate proteolytic cascades by inhibiting serine-type endopeptidases. The biological role and disease relevance remain largely uncharacterized, but genetic associations include spondyloepimetaphyseal dysplasia, Strudwick type. While related Kazal-type inhibitors are involved in tissue protection, inflammation, and tumor suppression, there is limited evidence establishing a prominent functional or therapeutic role for SPINK14 itself. Current datasets do not indicate approved drugs, clinical utility as a biomarker, or major safety concerns. SPINK14 is a valid gene/protein target but is currently best described as a putative extracellular serine protease inhibitor of the Kazal-type family.
Not applicable as no drugs have been described to target SPINK14. By analogy with other Kazal-type inhibitors, mechanism would likely involve competitive or allosteric inhibition of serine proteases.
