Serine peptidase inhibitor Kazal type 9 (SPINK9) is a cationic, Kazal-type serine protease inhibitor that is highly specific for kallikrein-related peptidase 5 (KLK5), a protease responsible for initiating skin desquamation. SPINK9 is predominantly expressed in the palmo-plantar epidermis, where it is thought to regulate skin differentiation and barrier function by inhibiting KLK5-mediated proteolysis. Additionally, SPINK9 can promote keratinocyte migration through a metalloprotease and EGFR-dependent mechanism. Pathological alterations in SPINK9 have been associated with certain skin diseases such as Norwegian scabies and pompholyx[1][2][4][5]. No drugs are reported to target SPINK9 directly.
