Overview

Serine protease 12 (PRSS12), also known as neurotrypsin, is a secreted trypsin-like serine protease primarily expressed in neurons and found at synapses. It contains domains characteristic of protein family S1, including a Kringle domain and scavenger receptor cysteine-rich regions. PRSS12 cleaves the heparan sulfate proteoglycan agrin, which is essential for the formation and maintenance of excitatory synapses. Its activity is linked to neuronal plasticity and learning; mutations cause an autosomal recessive form of intellectual disability, highlighting its importance in cognitive development[1][2][4][7].
