Serine protease 51 (PRSS51) is a predicted serine-type endopeptidase encoded on human chromosome 8. It shares homology with other testis-enriched serine proteases, particularly PRSS55; however, functional knockout studies in mice demonstrated that loss of Prss51 does not impair fertility or essential reproductive functions. PRSS51 is expressed at highest levels in the testis and liver, and is thought to reside in extracellular compartments. While its sequence conservation across mammals suggests evolutionary pressure, definitive biological roles—especially in human physiology or pathology—remain to be clarified. No compelling evidence supports its involvement in disease or its potential as a pharmacological target[1][2][3][4]. Key caveats: PRSS51 is frequently studied alongside PRSS55, which is critical for mouse sperm maturation, but PRSS51 itself appears dispensable for fertility and no non-redundant phenotype has been established in knockout models[1]. There is no evidence that PRSS51 currently qualifies as a valid drug target or disease biomarker, nor is there significant human disease implication based on available omics and pathway annotations[2][3].
